Predicted the attachment of conjugates to specific sites on MAbs through site specific cleavage of intact MAbs and analysis by mass spectrometry. ●Methods for binding specific antigens to affinity supports, affinity/immunoaffinity chromatography. ●Use of affinity binding techniques to recover ADCs from plasma to determine their structure during the PK cycle. ●Sequencing of peptides using the LTQ linear ion trap and MS2 and MS3 to confirm the sequence of residues in peptides. ●Development of bottom-up proteomics techniques to sequence antibody drug conjugates and determine the presence of metabolites in plasma. Specialties: AP Biosystems 4800+ MALDI-TOF, LTQ-FT, LC/MS/MS (triple quad, hybrid TOF, linear ion trap).
